The thyroid gland produces hormones that regulate metabolism, energy use, body temperature, heart rate, digestion, growth, and several other body functions. Thyroid peroxidase is an enzyme required for the production of the thyroid hormones thyroxine and triiodothyronine.

In an autoimmune thyroid disorder, the immune system mistakenly recognizes components of the thyroid gland as foreign. It may produce anti-TPO antibodies that target thyroid peroxidase and contribute to inflammation and progressive thyroid-cell damage.

Anti-TPO antibodies are commonly found in Hashimoto's thyroiditis, a major cause of hypothyroidism. Hashimoto's thyroiditis may initially produce few symptoms, but progressive thyroid damage can lead to fatigue, weight gain, cold intolerance, dry skin, constipation, hair thinning, menstrual changes, slow heart rate, and thyroid enlargement.
